Fingerprinting Arctic and North Atlantic Macroalgae with eDNA – Application and perspectives

Macroalgae are key primary producers in North Atlantic and Arctic coastal ecosystems, tracing their fate distribution is vital to improve our understanding of ecological role provision ecosystem services. Recent advances from environmental DNA (eDNA) have added a new capacity fingerprint trace macroalgae. However, further development resources for amplifying identifying macroalgal eDNA much needed. Here, we examined the performance terms resolution specificity two 18S primers (18S-V7 18S-V9) recently applied macroalgae eDNA. We also built local barcode database primer 18S-V7 with 31 widespread species complement existing databases. Furthermore, metabarcoding identify marine sediments (Disko Bay, W. Greenland) evaluated contributions database. identified 19 families across 11 orders surface (0–1 cm, both primers) sub-surface (5–10 primer) sediments. The developed here improved identification unique families, 16 thereby strengthening taxonomic assignment possible relative pre-existing reference sequences. Overall, this study demonstrates feasibility resolve sediments, enhances fingerprinting resolution. document novel pathway answer questions on Arctic.
